Very much so. -San is used as a sort of "Mr. or Mrs." in Japan, and is also used towards to show equality between you and the other.
-Kun is towards one to whom you want to show respect, more so than that of -san, it is also something that is often used when referring to boys.
-Sama means lord and is used when talking to someone who is much higher then you, it warrants for very high respect.

Also, thanks Phoenix, for some reason the word "arrogant" wasn't coming to mind, so I said big-headed.

actually kun is generally used when talking to somebody who is your junior, while san is used between equals. In some cases kun can be used by a girl who has a close bond with a boy; it's often used in anime this way, so non-japanese speakers tend to think that it is a sign of respect, but this is actually the less common way of using it.
