Figured it out, as far as I can.
Long story short:
Loading Nyx seems to do something, somehow, to patched EHCI drivers. This necessitates (sometimes multiple) reboots, and even re-patching the drivers.
The strange thing is that all other USB devices continue to function properly afterwards. Whatever happens to the drivers doesn't affect normal functionality, only breaks the ability to send payloads.
Worse, it's transient - I now have a boot on my laptop where I can semi-consistently launch Nyx... had to hotpatch the drivers twice, but have had about a dozen successful payload sends (trying to migrate to a new SD card... pain).
The electron gods must be chortling to themselves, or facepalming so hard that they have whiplash. >_>
